I have probably a complex sounding question.

Currently I am looking for a job as I have moved to a new area. I reached out to my network and got numbers for a few people to talk to.

One lady said she passed my info and CV on to her direct and he would get in touch with me tomorrow (a week ago). Do I chase this up or let it go?

I only have the number for the direct's boss and when I called her a 2nd time she said to wait out for the call from her direct.

Would calling again inform the lady that her direct is not doing what he is being told to or am I just being fobbed off?

Right there.  Do that.

Do *NOT*, under *ANY* circumstances, "inform the lady that her direct is not doing what he is being told".  This person is doing you a *favour* by passing your CV to her direct.  She's very unlikely to have given her direct an order to call you, and so the direct isn't "not doing what he is being told", but merely putting you into his giant pile of "stuff to do".

I strongly recommend that you "breathe in, breathe out, move on".  You should have any number of leads and opportunities on the go at the moment, so just put this one into the giant pile of "maybe", and work on other more promising leads.  That way, if you do get a call back, it's a pleasant surprise and you can deal with it with a positive, "Hey, great to hear from you!" attitude, rather than a negative "f**king FINALLY!" attitude, which I can assure you will come across to the other person and do nothing good for your candidacy.
